The Pinch Valve is located on the Vaccine Tube and is used as a barrier that allows/blocks the vaccine flow between the vaccine bottle and the Control Unit. 

It should be closed while switching to a new vaccine bottle or for any other reason you want to stop the vaccine from flowing.
When Priming, Calibrating, or vaccinating the pinch valve needs to be open so the vaccine can flow to the Control Unit.
